My question is about punctuating the introductory phrase, “This in turn” in the sentence “This in turn requires them to rely heavily on their volunteers to carry out their mission.”Can you tell me if two commas are needed – one after “this” and the other after “turn?”

Two commas are needed exactly where you suggested, because “in turn” is a nonrestrictive phrase. In plain English, this means it does not restrict the meaning of the sentence in any way, and you could omit it completely without changing the sentence’s sense.
